Okay, so there was a time when I was a little obsessed with WWE. And for whatever reason, I just remembered that. So, can you picture the guys in a setting like this? Doesn't even have to necessarily be wrestling, maybe more mixed martial arts. But there are cameras backstage and fights and lots of "entertainment". That training fight scene with Alec and Magnus is living rent free in my head and I would just like more of that, you know?

As it always does, my brain went right to "backstory" Mode. -

So, Alec was classicly trained, in a studio, with professional trainers and wrestling and other fighting styles have been practiced in his family for generations and are highly regarded. There is a philosophy to it, a feeling of honor. Alec's parents actually weren’t happy when their son went to the WWE (or MMAE) with his talents, but Alec argued that this might be able to help get the younger generation to look at the sport as something more than bashing each other for the entertainment value of the audience. (And, well, also money. Because while there is a familial wealth to the Lightwood name, Alec would like to be a bit more independent from them. And if he gets famous enough, gets his name out there, maybe he could open up an mma studio where he can then teach kids and adults alike the real sport.)

Magnus comes from a poor background and an even poorer neighborhood. If you didn’t know how to defend yourself, you were prey. When he was old enough to not be turned away by the guards immediately, he started fighting in underground rings for money. Fighting was never anything more for him than a means to make enough money to eat and survive. One night, a fighter manager looking for new talent happens to be at one of those underground rings and sees Magnus. He gets signed almost right away. And just months later, Magnus has money and a measure of security and is rising in fame. Without the worries of his past nibbling at his heels, he lives it up. Make it grand, make it a show, have some fun! Fighting was never fun before. It was survival. But now, Magnus lives.

Alec and Magnus would probably start out as kinda enemies, at least in Alec's mind. Because Alec thinks that Magnus is only in it for the show and the fame and the glory. That he has no respect for the Art as it is. And in a way, he is right. Magnus doesn't respect the art of fighting the way others in their line of work do. He didn’t learn it because he wanted to, but because he needed to. He will never feel the things that Alec does where fighting is concerned. But that doesn’t mean he doesn't have his own version of respect for it.

Once Alec understands that, he is even more in awe of Magnus than he was.

They both grow to learn and understand each other, without having to conform to the other's view. Magnus will never be able to see fighting as something inherently good, but he respects that Alec does. Alec will never see fighting as a mere tool, be he respects that Magnus does.

あと、522のピーク先からピンクリボンあって浅見茶屋への下へ下る道を勝手に誘導してたのもあるけど、それがヤバい。勿論急下りなんだけど、大木が完全に枯れ木で下がる時に軽く当たるだけでバキバキって木が倒れるという、危ない場所。 

水害でそのうち一気に流れるだろうね。

登山道が整備されてて明確な所なんかは作業道にピンクテープが付けてある事もある 

山を始めたばかりの頃、奥多摩でピンクテープ追いかけすぎて通せん棒無視してしまい作業道から廃集落に迷い混んでしまったのは良い思い出

以前に歩いた時のログ見たら見事に堂平山も六石も記録されてたわ。 

堂平の方は天覚山方面から歩いてくると方向的にも踏み跡的にも堂平山の方が道なりっぽく見える。 

六石の方はピークの手前左の方に巻き道っぽい踏み跡があるけど途中で消滅してた。

AbbVie’s Telisotuzumab Vedotin (Teliso-V) Demonstrated Encouraging Responses In C-MET NSCLC Patients Who Progressed On Previous Treatments

Phase II study of telisotuzumab vedotin in patients with c–MET+ NSCLC who have previously received not more than 2 lines prior systemic therapy (Abstract # 9016)

An anti-c-MET antibody-drug conjugate (ADC) called telisotuzumab vedotin is made up of a monoclonal antibody attached to monomethyl auristatin E. (MMAE). In the Phase I Study (MN14-237), Teliso-V and osimertinib are given to patients with previously treated NSCLC who have c-MET overexpression. Recently, the FDA granted the medication Breakthrough Therapy Designation (BTD) for use as monotherapy in patients with metastatic or advanced EGFR-wt non-squamous NSCLC whose cancer has progressed or grown after receiving platinum-based therapy.

Results of patients treated with Teliso-V and osimertinib in Phase I/Ib who had EGFR L858R, ex 19 deletions, and overexpression of c-MET. An ORR of 58 percent was seen when Teliso-V and osimertinib were combined. According to its recommended dosage, Teliso-V had an ORR of 3 (43%) out of 7 patients receiving 1.6 mg/kg and 8 (67%) out of 12 patients receiving 1.9 mg/kg.

A preliminary analysis from the Phase II LUMINOSITY trial, which involved 136 patients and of whom 122 could be evaluated for the main outcome, was given by AbbVie. The objective response rate (ORR) as determined by an impartial central review served as the primary endpoint. The Bayesian model predicts that stage 2 advancement is more likely if at least 70% of the genuine ORR is present. 131 (96%) of patients experienced treatment-emergent adverse events (TEAEs) of any grade, and 65 patients experienced Grade 3+ adverse events (48 percent of patients).

Conclusion

The effectiveness of both cytotoxic chemotherapy and targeted therapy is affected by the anti-cancer treatment class known as antibody-drug conjugates (ADCs), which can deliver cytotoxic medications directly to tumour cells. The use of ADCs is expanding quickly, and early research in lung cancer has produced encouraging results. ADCs that target HER2, HER3, TROP2, CEACAM5, and MET are now being tested in clinical studies for NSCLC.

Oncogene tyrosine kinase receptor (c-MET) is the term used to describe this substance. Numerous cellular processes can be regulated by c-MET, and when these processes are dysregulated, tumour cells are more likely to proliferate, survive, invade, and spread. Point mutations, amplification, fusion, and protein overexpression are c-MET changes found in NSCLC that are linked to a poor prognosis.

MET activation has previously been implicated as both a primary oncogenic driver mutation and a secondary driver of acquired resistance to targeted therapy in other genomic subpopulations, according to preclinical and clinical investigations. Agents that target c-MET are thus a possible therapeutic approach for NSCLC. There isn't currently a targeted medication that specifically targets NSCLC with c-MET overexpression. Therefore, obtaining a BTD can be essential in securing approval for the primary therapy in this patient group by providing this chemical.

okay tbh like the majority of comments I get on ao3 make my day. there's this one user, Mmae, who left a string of fabulous meta comments on a lot of my fics, talking about the little things they liked and pointing out lines and symbolism and stuff and it still makes me smile to think of.

someone once described my writing as "word painting" and said they felt like they were in english class because they were enjoying analyzing it so much. and someone else told me that my descriptions are the perfect amount of detail and vague so they worked really well to conjure up images and smells and I was like, fuck man, I'm gonna cry now because I struggle with describing scenery because holding a picture in my head is so fucking hard (three cheers for probably aphantasia ig) so I rely on emotions and broad strokes more than the details and I was like you mean it's working???

also just seeing writers I look up to in my comments? hooooly shit it's so cool. what a feeling. the first time virgil commented on my work I lost my shit for like thirty minutes.
