Last Wednesday the fire alarm downstairs went off for 4/5 ear splintering rounds at 1:30am then fell silent as if nothing happened. Definitely no fire/smoke/heat issues. I put it down to a fluke.
Half an hour ago it did The Beep right above my head. You know The Beep. That sound that means ‘change my batteries’ but mine are connected to the mains so I was like ‘oh.’
I used the website I used to find the plumber and asked for a callback but like...it did the chittery beep twice again at intervals. I knew it’d be like the beating of the telltale heart going off every once in a while
So I googled it.
Turns out likelihood is, the 9 volt battery used as a backup could need replacing.
I had ONE nine volt battery by pure fluke. I have a little box filled with batteries (all new) and they are you know, the usuals, AA’s and then some of the tiny round bastards. BUT THERE WAS ONE 9 VOLT. FOR REASONS I DON’T RECALL.
So I get out the stepladder (may I bring up to you at this point I have fallen arches that mean I am very very nervous of being even up on step stools due to 0 balance ability and 1000 incidents to go against faith in my ability) and I go to twist it off.
Nope.

It’s difficult as hell to get the battery out even though there’s a little ribbon tab, you know the type you pull on and typically the battery pops up and is easy to remove?
Fight with the thing. Finally get it out. Have to unattach it from the wires. One was easy, the other was NOT.
Fetch new battery from the stair I’d put it on. Attach it to connecting things and then realise the electrician who worked on it last PUT THE FUCKING BATTERY IN THE WRONG WAY, THAT’S WHY IT WAS SO HARD TO GET OUT. Put it back in the OTHER WAY this time and components and wires fit much easier.
Tighten the tiny screw on the battery compartment.
Stick it back up and twist so it’s back on the ceiling.
Stand utterly anxious to the strands of my DNA in case it suddenly goes off or returns to its chirps.
Nothing. Did I do it?
Literally stood under the fire alarm and said aloud to utterly no-one
DID I DO IT?!
Sit down a moment, hands are shaking like I’m sitting through a high level earthquake and casting glances of supreme concern to alarm.
....It has not chirped at me again. It hasn’t gone off and scared me to death either.

I might have done it!

A common variant of CNTNAP2 is associated with sub-threshold autistic traits and intellectual disability

ABSTRACT
Sub-threshold autistic traits are common in the general population. Children with sub-threshold autistic traits have difficulties with social adaptation. Contactin-associated protein-like 2 (CNTNAP2) is associated with the development of Autism spectrum disorder (ASD) and the single-nucleotide polymorphism rs2710102 (G/A) of CNTNAP2 is suggested to contribute to sub-threshold social impairments and intellectual disabilities. We recruited 67 children with Autistic disorder (AD) (49 boys, 18 girls, aged 38-98 months) and 57 typically developing (TD) children (34 boys, 23 girls, aged 53-90 months). We assessed the participants' intelligence and social reciprocity using the Kaufman Assessment Battery for Children (K-ABC) and the Social Responsiveness Scale (SRS), respectively. Genomic DNA was extracted from the buccal mucosa and genotyped for rs2710102. A chi-square test revealed a significant association between genotype and group [χ2(2) = 6.56, p = 0.038]. When a co-dominant model was assumed, the results from linear regression models demonstrated that TD children with A-carriers (AA + AG) presented higher SRS T-scores [t(55) = 2.11, p = 0.039] and lower simultaneous processing scale scores of K-ABC [t(55) = -2.19, p = 0.032] than those with GG homozygotes. These associations were not significant in children with ASD. TD children with the rs2710102 A-allele may have more sub-threshold autistic traits than those with GG homozygotes, reflected in higher SRS scores and lower simultaneous processing scale scores. These results support the use of genetic evidence to detect sub-threshold autistic traits.

FDA Gives Inovio Green Light for Phase 3 Trial of DNA COVID-19 Vaccine ~ November 11, 2021 🧬🧬🧬🌎🌎🌎
Inovio ($ino) has received the FDA go-ahead for a phase 3 trial of INO-4800, a DNA plasmid vaccine against COVID-19.
The agency lifted a prior clinical hold on the study after Inovio satisfied questions about the vaccine and its needleless electroporation delivery system, Inovio’s Joseph Kim said on an investor webcast.
Inovio and partner Advaccine Biopharmaceuticals Suzhou Co. (China) will conduct the INNOVATE study in Mexico, Brazil, Columbia and Thailand, in addition to the U.S. The trial will evaluate INO-4800 in healthy adults aged 18 years and older, who will get two 2-mg doses given one month apart. The primary endpoint is virologically confirmed symptomatic COVID-19.
“We are aiming to have the interim phase 3 data available by the first half of 2022,” after which Inovio will apply for an Emergency Use Authorization, Kim said.
This is the first large-scale test of a DNA plasmid vaccine for COVID-19. DNA vaccines have some unique advantages over mRNA vaccines, including a relative insensitivity to temperature. The vaccines have shown 1-year stability at temperatures of up to 98.6 degrees and have a projected 5-year shelf life at normal refrigeration temperatures.
The company’s proprietary delivery device is a key component of the vaccine system. The hand-held unit, powered by AA batteries, uses air pressure to force the vaccine into the epidermis. It then generates millisecond-duration electrical pulses that temporarily open pores in all cells lying within the electrical field, including immune cells. This allows the plasmid ring, encoded with the viral antigen, to enter easily. Electroporation increases the amount of DNA delivery by more than 1,000-fold compared to traditional injection delivery, the company said.
After uptake, immune cells use the plasmid DNA sequence to generate antigens; this stimulates an immediate immune response as well as the long-lasting humeral immune memory vaccines rely on. DNA vaccines do not interfere with or change in any way an individual’s own DNA, the company added.
“The CELLECTRA 3PSP device is designed specifically for a COVID-19 type pandemic scenario,” the company said in a press statement. The user-friendly design “allows for stockpiling of the device in quantity without maintenance … [It] has been designed with reliability, challenging environments, user needs and ease of large-scale manufacturing in mind.”
Development of this device really took off in 2019, with an $8.1 million investment from the medical arm of the U.S. Defense Threat Reduction Agency’s Medical Chemical, Biological, Radiological and Nuclear Defense Consortium.
The lifting of FDA’s clinical hold is another in a series of green lights flashing for Inovio, Kim said on the webcast.

Roland MC-101 review: Beats, bass and batteries
If you think Roland's nostalgia-infused music gear is a new thing, think again. As far back as the mid-'90s (and maybe beyond), the company was cashing in on the gravitas of some of its legendary instruments. In 1996, the company introduced the MC-303 groovebox. It might have had "303" in the name, but the similarities with the iconic TB-303 pretty much stopped at the color scheme. The real news with the MC-303 was the birth of the beloved groovebox series -- all-in-one music-production machines.
The latest in that line is the MC-101, and it's small, somewhat affordable ($500) and surprisingly powerful -- and a welcome addition to the oft-forgotten family. Notably, it doesn't trade in nostalgia (at least, not in the way the MC-303 did), but it should still appeal to music enthusiasts who love Roland's classic music boxes.
Before we get to what it does and how it sounds, the main selling point here is clear: portability. At just 9 x 5 x 2 inches and a shade more than 700 grams (1lb, 9oz), the MC-101 is easy to bring with you anywhere. Add in the ability to run it on four AA batteries (or USB power) and it's clear that this was meant to move with you. And for those who prefer to stick to the studio, the MC-101 should fit into almost any small patch of free desk space.
If the MC-101 looks familiar, that's because it has a fairly common groovebox layout. But the MC-101 is particularly similar to the much bigger, pricier Roland MC-707 ($999). For good reason, the two share a lot of DNA: The MC-101 is basically the MC-707 mini.
Whereas the MC-707 is a fully fledged piece of studio gear, the MC-101 omits many of the bells and whistles, leaving you with the essentials for making tracks on the go. This means that around the back, there's not too much going on. There's a power switch, an SD card slot, two five-pin MIDI ports and a pair of ¼-inch outputs (the MC-707 has more output options, including send, return and "assignable"). The 101 also has a 3.5mm headphone jack on the front, unlike the ¼-inch one on the 707.
As for the main interface, there are 16 pads, four-channel faders and four rotary knobs -- no prizes for guessing that the MC-101 offers four tracks to play with. The rest of the controls relate to transport (play, record, et cetera), pad modes (clip, note et cetera) and general utilities -- like headphone volume or the multipurpose Value knob, which you'll be using a lot.
Both the MC-101 and the MC-707 come loaded with more than 3,500 preset sounds. These are organized into logical subcategories such as drums, bass and leads and then more specific sounds like piano, clav and wind (there are many more categories to explore). The range of sounds is comprehensive but skews toward traditional musical instruments rather than unnatural synth sounds. There are some good synth-based presets for sure, but if EDM or dance music is your thing (it is mine), you have to hunt a bit and be willing to experiment (more on this later) for something a little more rave-y.
Don't worry though, as you can load a sample of anything you want, which basically blows away any concerns about this thing not having the sound you need. However, that means having access to a library of sounds (either a sample collection or a synth to make your own) and then having to import them. It's not difficult to transfer sounds; mostly it's a case of adding them to a folder on an SD card. But if you like to browse different sounds as you have a melody playing, you need to plan ahead and have suitable samples at the ready.
For drums, there are the usual predefined kits, including classics from the TR-808 and TR-909, along with plenty of alternatives (live/natural drums, jazz kits and so on). I was surprised at the lack of TB-303 lead sounds though. I know it's been overdone and it's kind of hard to capture the wonky magic of the 303 in a sample, but I was hoping there'd at least be some of that squelchy goodness. Instead, there are maybe two or three 303 sounds, and none of them are of the acid-house variety (which is what most people might be hoping for).
As with all grooveboxes, there's a learning curve, and it's moderately steep. The MC-101 isn't difficult to use, but it can feel a little overwhelming at first. The problem with these devices is that they all have their own distinct workflow, so it feels like starting over every time. With practice, I started to realize how Roland wants you to work. Some things are obvious: Pressing Project will open the project menu on the tiny, text-only display. Rotating that Value knob cycles through the options (Load, Write, New, et cetera), and you'll click that rotary to select.
Some things are a little more hidden. There's a prominently placed Shift button on the top left of the MC-101. Anything beyond basic note input is generally accessed with Shift plus... well, almost every knob and button has a second function, and often more. The confusion set in when I was trying to find a setting that I knew I was doing the right key combination for but hadn't realized I was in a different mode (say, Clip mode, rather than Track mode) and it would drive me crazy.
This gets worse when there are other variables in the mix. For example, track one is basically dedicated to drums whereas other tracks can be assigned to Tone (i.e., an instrument) or Looper (better for performances). I spent a few minutes trying to create a Tone track, forgetting I was on track 1. These confusing moments mostly disappear once you have the lay of the land -- although I am still finding more menus and options every day.
Effects, in particular, aren't the most intuitive. If you want to add a little crunch to your kick drums, for example (and, please do!), then you have to press Shift+Sound and then navigate a scroll menu on-screen, click into the effects section and rotate through the available options to find the one you want. Then you have to back out and choose another menu to alter its parameters. Not a huge chore, but it's a few more steps than usual. For what it's worth, the selection of effects, once you do find them, is generous. I started to count but stopped. Roland's website says there are 90, and I'll have to trust it. So whether you're looking for distortion, chorus or delays, you're probably covered.
Sadly, the same can't be said for sound-shaping options. This is one of the main differences between the MC-707 and the MC-101. On the 707, there's a larger, graphical display, and you can dial right down to the individual oscillators. With the 101, you have far fewer controls, although the ones you do have are most of the ones you need (envelopes, cutoff, resonance and so on). I would have loved to have had the option to change wave forms and detune, but there's enough here to keep you going.
Once you have the sounds you want and your tracks set up, sequencing is a pretty standard affair. As with most hardware, you can step sequence or record live, depending on your preference. I kinda suck at "playing" (even with quantize to help) so tend to lean toward step sequencing. These are grooveboxes after all, and step sequencing is perfect for "groove"-based music. Whichever method you prefer, it's easy enough to extend the length of a clip to eight measures. Page (arrow) buttons on the left let you see the notes in play and edit after the fact.
In 2019, having only four tracks to create a song with might seem a little stingy. Some of my DAW productions have four tracks just for different versions of a kick sample. Despite this limitation, the MC-101 has a few tricks up its sleeve. Most obvious is Clip mode. The terminology here makes it difficult to avoid repeating myself, but I'll give it a shot.
Under each of the four tracks, you can build a clip (a phrase, melody or rhythm, depending on what you're doing). You can create new clips on the same track, one per pad, which you can then trigger to play in any order you choose, allowing you to build up a drum beat or melody over time so that your song has progression. If your track is set to Clip mode, you can even change the instrument/sound on a per-clip basis (say you want a sax solo to become a sawtooth lead after eight bars). With this, you can easily make several variations on an idea with different chord progressions, lengths and instruments all within one "track."
The result is that with a little bit of creativity (and button mashing), I can create a song with a number of different components, which I can then order in any way I choose -- or even trigger in a performance, much like Ableton Live's Session view. Throw in the fact that there's a Motion Designer (what appears to be Roland's name for automation) and it's not hard to see how you can go beyond simple loops strung together for a few minutes to something that's dynamic and fluid.
In fact, the more I use the MC-101, the more capable it seems to become. I've always felt that grooveboxes, as fun as they are, were primarily idea machines that you then want to export over to a DAW for fine-tuning and completion. But over the years they've become more adept as complete song-making tools in their own right, and even the plucky little MC-101 has a lot of detailed tools to keep your grooves from feeling, well, boxy.
If I had any negatives, it would be some of the more practical stuff. First of all, there's the battery life. The MC-101 ships with four AA batteries, but they only lasted a couple of hours. Roland claims the battery life is around three hours for Alkalines (that's what came in the box) or up to five for Ni-MH cells. If that five-hour claim holds true, it's decent enough, but it's something to be mindful of.
I'd even say that having to use AA batteries is a negative. I generally dislike single-use batteries, as they're wasteful and need recycling. I have good rechargeables, but the life on them is generally worse, so there's a trade-off. I would suggest that a built-in battery might be a better option, but a benefit of AAs is that you can swap them out quickly and be back on your project in no time. That said, you can technically run this off a power brick as long as you have the right USB cable (Type B, not included), but that feels a little clunky. You can, of course, just run it off USB indefinitely if you're at home.
Another slight annoyance is the headphone volume: It's adequate at best. It'll depend on the impedance of your headphones, but chances are, if you're serious about your productions, you have a fairly good (and probably, therefore, high impedance) set. When I switched to a pair of cheap buds, volume improved but at the expense of the sound quality. You can, of course, connect this to an audio interface, but that's less helpful if you're on the move.
Overall, if you're looking for something portable, versatile and musically capable, Roland's MC-101 is exactly that. While you might get more control over your sounds from Korg's cheaper Volca line, those aren't all-in-one production tools. Teenage Engineering's OP-Z is a worthy rival, depending on your music preference and workflow. It offers more tracks (16) and has some truly novel features, but it's also slightly harder to work with and has a few quirky flourishes that make it more of a specialist device. It's similarly priced to the MC-101 though, so it's definitely worth a look.
For those looking for a good all-round music-making machine that will fit into your studio (and your backpack) and that boasts a library of very usable sounds, can play samples and is big enough to perform with, then the MC-101 might just well be that.

"Where is he anyway? He hasn't been by since left the house this morning. Do you think he's trying to sext you from the grocery store again?"
"Just go, I'm ready to kick your Batteryman ass."
Kajii grinned and obliged, taking a card from his hand and raising it dramatically.
"Well, since I have three or more Batterymen on the field, I'll activate—"
Chuuya waved his hand.
“Oh no, you've had three Batterymen for the past 2 turns, and you've been playing what you draw, have you just been hanging onto that card just for the hell of it?"
Kajii practically glowed as he set Short Circuit onto to field.
"I drew it in my starting hand, my dear lemondrop; it's much more fun when your field has more monsters. Can you counter it?"
"No, but do you realise you don't have to play like a jackass?"
"I like to watch your reactions. Each of your defeats is a valuable sample."
Chuuya gave him a look and put his four monsters in the graveyard.
Kajii moved to continue his turn and go in for the kill.
"Now I'm going to—"
Chuuya laughed.
"Attack directly for 9000? Yeah, no; I activate Threatening Roar."
"Then I pay 1000 Life Points to activate 7 Tools to negate it!" Kajii said, dramatically flipping a trap of his own.
But Chuuya was ready.
"Then bitch, so do I!"
He said with a grin, flipping up a second trap card triumphantly.
Kajii frowned. He wouldn't be able to attack that turn after all.
Not that he was particularly worried about it. It'd be a little more difficult, and he wouldn't get the really fun OTK, but he was still set up well and Chuuya's new Machine deck was only sort-of holding together. It wasn't anything flashy.
"Fine." He conceded. "I end my turn. We both pay 1000; I have 5250 and you're at 4500."
Chuuya's phone went off again.
"BITCH! I'm playing Yugioh!" He said loudly at the screen, as he read Dazai's message, his nails audibly tapping around as he typed his response. Kajii smiled softly. Chuuya was so...expressive.
"Tell him to come over." Kajii suggested. But Chuuya was already drawing a card from his deck to start his turn.
"I told him to just jack off." The redhead said lightheartedly. A smirk grew on his face. "Though he really should be here to watch me finally hand your ass to you."
"Ooh, cocky banter, is this where I'm supposed to be concerned?"
"Tch."
Chuuya placed a card on the field, and then another.
"Since I don't control a monster, and you do, I can special summon Cyber Dragon from my hand. Then, for my normal summon, I'll play Biofalcon."
Kajii pretended to not be listening.
"Did you know that you can make batteries from lemons? I can show you after I use these batteries to win again."
"Oh, shut the fuck up, I activate DNA Surgery and—"
Suddenly Kajii understood the purpose of Chuuya's deck.
"Oh no, you won't, I use 7 Tools—"
Kajii stopped mid-sentence and looked down at his Spell and Trap Zone, cursing under his breath. He'd used it already.
Chuuya's grin stretched across his entire face.
"Can't negate?" He asked with a smirk.
Kajii just looked at him with dread, and Chuuya continued.
"In that case, I activate DNA Surgery and I call Machine. All monsters on the field are now Machine monsters."
Kajii's ego was dropping rapidly and was fast on its way to the molten core of the Earth.
"You have one of those don't you?" He said with resignation, watching Chuuya's face. The redhead looked on top of the world as he pulled a card from his Extra Deck.
"If by 'one of those' you mean a Chimeratech Fortress Dragon, fused from every monster on the field, that's attacking you directly for 6000, and therefore for game? Yeah, I have one."
Kajii clapped slowly.
"You win, good job, babe."
Chuuya smiled happily as he picked up his cards and shuffled his deck. He was really cute, and Kajii wondered how many successive victories it would take for his reactions to shift. He picked up his own deck and put it away.
"Finally." He said, collapsing backward on the bed.
Kajii picked up his cards and laid down beside him, playing with a strand of his hair.
"That was a great play." He praised.
Chuuya chuckled.
"Someone had to do away with those batteries."
"Then I'll just have to find a new build to test you with. The pursuit of science never ends."
"What the hell sort of science are you doing anyway?"
Kajii chuckled, enjoying the idle comfort of lying with Chuuya, of his hair between his fingers and his small frame beside his. He didn't answer Chuuya's question. The results would be tainted if Chuuya knew the details of the study.
Kajii looked down at Chuuya's face. A satisfied smile was still present on his lips. His eyes were closed, and he looked comfortable and happy.
A knock came from the door.
"Can you get that, Motojirou?" Chuuya drawled. Kajii nodded and kissed his forehead softly before pulling himself off the bed and making his way to the door of the apartment.
When he opened the door, he found Dazai standing outside, a large pizza and tray of cookies in his hands, and a grocery bag hanging on his arm.
"Hello, Motojirou!" He said, his voice practically singing. A kind smile was on his face. "Did Chuuya win yet? I brought things to celebrate."
Kajii smiled, opening the door wider for Dazai to get in.
"He won about ten minutes ago," Kajii said as Dazai came in and started toward the bedroom, Kajii following behind him. "How'd you know he'd win?"
Dazai's foresight actually regularly amazed him. He was nightmare to play with, he was great at predicting what people would do, and he ran lockdown decks that had to have come straight out of hell itself. You couldn't do anything. You couldn't win.
"Chuuya and I worked quite a bit together on that Machine deck. It was tailored just for him, so I had a feeling."
Dazai set the food down on bed.
"Hey, love." He said, giving Chuuya a gentle smile. Chuuya pulled himself to a sitting position.
"Oh, hey, aren't you supposed to be beating off in a public bathroom?" He teased.
Dazai batted his hand.
"Not if the alternative is celebrating with you."
He said it sweetly, as if that could save the fact that he'd basically admitted that he would be otherwise.
Chuuya rolled his eyes, and Kajii and Dazai both joined him on the bed, kissing the top of his head together.
They'd had this sort of relationship for a couple months now. Kajii didn't know Dazai terribly well, but he and Chuuya had a lot of history in the past. And Kajii had his own relationship with Chuuya. And Chuuya loved them both.
Kajii had been a little jealous of Dazai at first, but he figured out quickly that Dazai wasn't competition. They weren't vying for affections, they merely shared them. And they got to make Chuuya smile together, and trap him in hugs, and hold both of his hands. He might not feel what Chuuya felt toward Dazai, but he'd decided that he didn't really need to. Dazai was his friend now, and perhaps in time he'd grow on him romantically. Even if he didn't; the relationship they had now was wonderful. It didn't need to change.
"How badly did he beat you, Motojirou?" Dazai asked, pulling a slice of pizza from the box.
"I lost with -750." He answered, taking a slice of his own.
Chuuya smiled smugly.
"I took all of his AAs and a Charger for Chimeratech." He added.
"Nice!" Dazai said, but his congratulatory expression disappeared as Chuuya made to grab his pizza.
"Get your own, pipsqueak." He said with a teasing laugh, holding it out of reach. "There's a whole box."
Chuuya huffed.
"I bet 'Jirou'd give me his." He pouted.
"Of course, lemondrop."
Kajii took another slice from the box and handed his to Chuuya, smiling and earning himself a kiss on the cheek. He gave a small giddy laugh.
He had everything he could want.
And with the way the night had gone, with Chuuya here, with Dazai here...with the game they'd played, and the expressions his boyfriend made, and the kisses he'd given and gotten —as far as he was concerned, that was all he needed.
He was happy.

Mail Forwarding: Workhorse Vies for USPS Contract, Readies Vans, Drones, Copters

The Workhorse N-Gen sounds at first description like a Hollywood spy-movie cliché: a near silent electric powertrain within a white van with a rather frumpy, nondescript exterior—and a little drone aircraft deployed from the roof to make the final move to the target.

Its reason for being is much more pragmatic, however; it’s the latest in a growing line of delivery vehicles from Workhorse of Ohio, positioned for the greening of corporate fleets while keeping ownership costs extremely low. The N-Gen is designed to replace a generation of small delivery vans powered by gasoline and diesel engines. Although final specs aren’t out yet, it’s expected to weigh hundreds of pounds less than those older vans, thanks to a lightweight composite body. Electric motors will provide about 100 miles of plug-in power, while a small onboard gasoline-fired range-extending engine will add another 75 miles of range. Workhorse claims up to 65 miles per gallon equivalent (MPGe) for the N-Gen.

Although Workhorse has been teasing its W-15 range-extended electric pickup (pictured below), which we briefly drove earlier this year, for much longer, it’s the N-Gen that will reach the market first; production will start in the first quarter of next year. “It’s coming out first because regulatory-wise it’s just easier,” explained CEO Stephen Burns, explaining that it’s closely related to the vehicle that’s a finalist in the United States Postal Service Next Generation Delivery Vehicles (NGDV) vetting process. Workhorse, in conjunction with truck outfitter VT Hackney, delivered its six prototype vehicles on time in September for evaluation. The USPS is expected to announce around March 1 what company gets the contract to build up to 180,000 vehicles over a time period of up to seven years.

Burns said that with the potential for high-volume production (and long-term support), lining up Tier 1 suppliers for the USPS project hasn’t been an issue. So it was a natural next step to develop a commercial-van version at the same time.

Panasonic (Tesla) Batteries, BMW Range Extender

The N-Gen and Workhorse’s W-15 pickup will share quite a lot, including their composite body construction. Burns told Car and Driver that they’ll use the same batteries: Panasonic 18650-format cells, a “commodity” format that is roughly AA size and essentially the same as is used in the Tesla Model S and Model X, among other vehicles. But what’s different is the range extender. While the production version of the W-15 will use a 1.5-liter three-cylinder BMW range extender, the N-Gen is going to use the 647-cc two-cylinder Kymco-built engine from the BMW i3, Burns said.

That’s because, while the van would only be aimed at commercial use, with well-defined driving loops and the range extender strictly as an emergency crutch, the pickup is being designed with a wider operating range—including sustaining its peak power after the battery is depleted.

Burns said the general public has given the electric pickup an unexpectedly strong reception, and now the company is trying to decide whether to build a consumer-focused version. “We’ve really been wrestling with it,” he said. “But we want to cut our teeth with fleets because that’s our DNA. We want to make sure we never disappoint them.”

The N-Gen has all-wheel drive, which is unusual for a commercial van. “These delivery guys have to go in all sorts of weather,” Burns said. Meanwhile, the company touts the low-floor design combined with the body-on-frame layout and standard pickup-truck ride height as providing a good blend of clearance, durability, and loading ease.

Workhorse says that its electric vehicles have already logged nearly two million miles and are in use in 14 states. The company has been ramping up production at an Indiana plant; this summer and fall, it made 143 of its larger E-Gen trucks for the United Parcel Service (out of a total order of 200 from UPS), and it is currently at a production rate of about three vehicles a day. W.B. Mason has also placed an order for those larger vehicles, Ryder is providing sales and support, and Workhorse has one other large order pending that Burns can’t yet talk about. With its current factory, Workhorse can make 60,000 vehicles of the roadgoing kind per year. And if the USPS contract happens? “We could probably fit more,” Burns added. “But it falls under ‘good problems to have.’ ”

It’s Really about the Last Mile

If you’re in the shipping logistics business—or if you have anything to do with e-commerce and shipping goods—“last-mile delivery” is what it’s all about. In the last mile or few miles of getting a parcel to its final destination, the costs ratchet up, and the task becomes more complex. It’s exactly what Workhorse appears to be trying to address with its approach, which embraces electrification and, when necessary, takes to the skies. According to a report last year from McKinsey & Company, 60 percent of consumers are either in favor of or indifferent to drone delivery.

Workhorse hasn’t yet said much about the HorseFly drone that comes with the N-Gen, but it’s a model that the company engineered in-house. It will deploy from the vehicle and will be software linked, so that it might be able to take a small package to a nearby cul-de-sac, for instance, while a larger one is being delivered.

This isn’t the only company vying for the last-mile delivery business and its electric future. One particularly well-funded effort is Chanje, a startup that has managed to recruit former executives from Volkswagen, Tesla, and one earlier fleet-focused company, Smith Electric Vehicles.

And a Hybrid Helicopter for Two

Workhorse also has a larger, manned aircraft under development. The unique eight-blade SureFly Octocopter, which the company revealed at the Paris Air Show this summer, can hold the pilot plus one other occupant. It’s also an electrified product, but with power sources prioritized the opposite way compared to its trucks; there, a gasoline engine provides power to the electrically driven prop system, and twin battery packs serve as a five-minute power backup, allowing the pilot enough time to land if they run out of fuel or the engine fails.

Burns said that the company is hoping for its first manned flight of the vehicle toward the end of the year or the first week of January and hinted that there may be a first public demo at the 2018 CES technology show in early January.

Considering Workhorse’s wide portfolio of intended products, from larger delivery trucks to vans and pickups and from copters to drones, what comes as a complete surprise is that the company operates in the minor margins. A third-quarter financial report this last week listed total operating expenses, including R&D, of $8.4 million for the quarter. That’s small change; by comparison, Tesla’s R&D spending in the same quarter was at about $332 million, and BMW’s was $4.1 billion. Burns agrees that while they’ve done a lot with a little, they need a partner to see the full vision through.

Amazon Prime Day 2017 Tops Black Friday and Cyber Monday
Amazon is thrilled that its just-wrapped Prime Day sales event was its biggest global sales event to date. The shopping extravaganza surpassed its Black Friday and Cyber Monday tallies, combined.
According to Bloomberg News, “The 30-hour sale, featuring discounts on a broad assortment of goods, generated about $1 billion in revenue for the Seattle-based web retailer, according to separate estimates from analysts at Cowen & Co. and JPMorgan Chase & Co. That’s triple its average daily e-commerce revenue in 2016, according to data compiled by Bloomberg.”
And more importantly for Amazon, it boosted its Prime subscribers and sold more Echo devices to draw more customers into its ecosystem.
The Prime Day 2017 event grew by more than 60 percent over last year. Sales growth by small businesses and entrepreneurs was even higher, with some reporting their own booming Prime Day sales results.

Inovio Receives $5 Million Grant from Bill and Melinda Gates Foundation for COVID-19 Vaccine Program
In a recent press release, Pennsylvania-based American biotechnology company Inovio Pharmaceuticals (Nasdaq: INO) reported that the Bill and Melinda Gates Foundation has provided a $5 million grant to support the accelerated evaluation and production of their "CELLECTRA 3PSP" medical device, which is integral to the intradermal delivery of their DNA-based COVID-19 vaccine INO-4800. INO-4800 is currently being tested in preclinical studies and is anticipated to progress to Phase 1 clinical trials in the United States during April 2020.
President and CEO of Inovio Dr. J. Joseph Kim stated, "INOVIO is grateful to the Bill & Melinda Gates Foundation for their continued investment in INOVIO's DNA medicines platform and for their support for DNA vaccines to potentially protect those at risk globally given the current COVID-19 outbreak. Our team of vaccine experts are working around the clock to advance INO-4800 and we look forward to attracting additional partnerships to expedite its development to meet this urgent global health need."
According to the press release, "The next generation CELLECTRA 3PSP device is designed specifically for a COVID-19 type pandemic scenario. It is a small, portable, hand-held, user-friendly device that runs on readily available "AA" batteries. This allows for stockpiling of the device in quantity without maintenance. It is easy to use and is based on our current device with extensive history (over 6,000 administrations) which has received the CE mark and has an acceptable safety profile. The streamlined design also allows it to be readily produced at reduced costs and large scale."
"The device has been designed with reliability, challenging environments, user needs and ease of large scale manufacturing in mind. INOVIO's San Diego Device Manufacturing facility will build initial quantities and demonstrate the design and scale up of manufacturing processes which can then be transferred to additional contract manufacturers for increased capacity. Initial development of CELLECTRA 3PSP was started in 2019 with $8.1 million funding from the medical arm of the U.S. Defense Threat Reduction Agency (DTRA)'s Medical CBRN Defense Consortium. The new funding will help to accelerate the testing and completion of the device development and scale up to combat the COVID-19 disease," the press release stated.
Inovio intends to deliver one million doses of the INO-4800 vaccine by the end of 2020, and "is working to scale up both INO-4800 and CELLECTRA 3PSP devices to potentially make available millions of doses to combat this outbreak." The company has announced plans to start US-based human clinical trials of INO-4800 in April. During a meeting with the US Coronavirus Task Force on March 2nd at the White House, Inovio President and CEO Dr. J. Joseph Kim stated, "Inovio is the leader in coronavirus vaccine development and the only company with a Phase 2 vaccine for a related coronavirus that causes Middle East Respiratory Syndrome (MERS). On January 23rd, 2020, the Coalition for Epidemic Preparedness Innovations (CEPI) issued Inovio a grant of up to $9 million to fund preclinical and clinical development of INO-4800. CEPI CEO Richard Hatchett stated, "Given the rapid global spread of the 2019-nCoV virus the world needs to act quickly and in unity to tackle this disease. Our intention with this work is to leverage our work with Inovio on the MERS coronavirus and rapid response platform to speed up vaccine development."
Dr. Kim responded, "We're extremely honored to expand our partnership with CEPI to tackle this new threat to global public health. Our DNA medicine platform represents the best modern day approach to combatting emerging pandemics. We have already demonstrated positive clinical outcomes with our vaccine against MERS-CoV, another coronavirus. Importantly, following the Zika viral infection outbreak, Inovio and our partners developed a vaccine that went from bench to human testing in just seven months – the fastest vaccine development on record in recent decades. We believe we can further improve upon this accelerated timeline to meet the current challenge of the emerging Chinese coronavirus 2019-nCoV."
On January 30th, 2020, Inovio announced that it would develop INO-4800 in collaboration with China-based Beijing Advaccine Biotechnology Co., with the goal of expediting the testing of INO-4800. Dr. Kim stated, "Our collaboration with Beijing Advaccine and its Founder, Emeritus Professor Bin Wang from the prestigious Fudan University and China's premier DNA vaccine expert, will tremendously accelerate our coronavirus vaccine INO-4800 development in China because of its expertise and experience with regulatory authorities and clinical trial management. This collaboration allows us to enter China and deliver our vaccine into the areas where they need it most as soon as possible. Our shared goal is to utilize both company's expertise in developing vaccines for emerging infectious diseases and hopefully achieve an accelerated regulatory approval for INO-4800."
Since January, Inovio Pharmaceuticals has demonstrated immune responses to their vaccine in animal models and initiated plans for a human clinical trial. The company intends to finalize their clinical trial design in March and prepare 3,000 doses of their vaccine for trials in the United States, South Korea, and China. The first clinical trial will include 30 healthy volunteers in the United States, with the Asia-based trials occurring afterwards.
Inovio's DNA-based vaccine platform is centered around speed of manufacturing, stability during transport and storage, and efficacy for inducing a robust immune response. INO-4800 consists of small circles of engineered DNA that can be directly injected into cells through the controlled use of electricity, which creates temporary pores in the membranes of target cells and allows the double-stranded DNA circles contained within the vaccine to enter cells, alter their activity, and eventually lead to an immune response. Since Inovio's formulation is composed only of highly purified DNA, water, and salt, it avoids potential toxicity and can be manufactured relatively quickly and stored and transported without freezing.
During the epidemic of the Middle East Respiratory Syndrome (MERS), a coronavirus related to the virus that causes COVID-19, Inovio was the first biotechnology company to test a vaccine against the MERS virus (INO-4700) in humans. The company plans to further evaluate INO-4700 in a Phase 2 clinical trial in the Middle East, where the majority of MERS cases have occurred.

The results of a Phase 1 clinical trial evaluating the immunogenicity, safety, and tolerability of the MERS coronavirus vaccine GLS-5300 (INO-4700), published in the journal Lancet Infectious Diseases, demonstrated that their DNA-based vaccine technology is well-tolerated and capable of inducing an antiviral immune response. After two vaccinations, 47 of 66 (71%) study participants showed evidence of a T-cell response. 44 of 58 (76%) participants demonstrated a T-cell response after three vaccinations. Seroconversion was detected by S1-ELISA in 59 of 69 (86%) and 61 of 65 (94%) participants after two and three vaccinations, respectively, and 34 of 68 (50%) individuals were found to bear neutralizing antibodies. No serious adverse events were associated with the use of the MERS coronavirus vaccine. The authors stated that the antibody and cellular immune responses associated with the vaccine were durable and similar to those of patients that recovered naturally from infection by the MERS coronavirus.

The LifeStraw is the original straw-style water filter designed for hikers, bikers, and other people that enjoy the outdoors. It removes 99.999% of bacteria, parasites, and microplastics.

Do LifeStraws really work?
LifeStraws work extremely well. They remove more than 99% of all bacteria and parasites. This means it takes out 99.999999% of waterborne bacteria (this includes e.coli and salmonella) and 99.999% of waterborne parasites (including giardia and cryptosporidium).
It also removes all but the smallest microplastics and sediment.
Can you drink any water with the LifeStraw?
LifeStraws are a filter so they remove the sediment, bacteria, and protozoa from the water before you drink it. This means that you can grab a gross, dirty glass of water and just drink it through the LifeStraw without a problem. LifeStraws don’t filter fuel and other chemicals out of the water so you need to be sure that there isn’t any chemical contamination in the water that you plan to drink.
They also don’t remove viruses. If you suspect that the water has viruses of some kind in it, then you need to go with some other method of getting it clean enough to drink. If this is the case you can boil the water before drinking it through the LifeStraw. This will kill the viruses (as well as the protozoa and bacteria).
It also will not filter out salt from water. So while it may remove the bacteria and any parasites from saltwater, you still can’t safely drink it unless you remove the salt some other way.
How many times can you use a LifeStraw?
The long life of the LifeStraw is one of its best selling points! You can filter about 1,000 gallons of water with one LifeStraw. That’s about 5 years worth of water.

Do LifeStraws remove viruses?
LifeStraws can’t remove viruses. Viruses are too small to be filtered out by most filters including the LifeStraw.
How do you clean a LifeStraw filter?
LifeStraws can’t be broken down and cleaned like some other water filters. The best that you can do is to blow out the LifeStraw after each use and leave both ends open so the filter can dry out.
You can also suck some clean water up into the filter and then blow it back out. Of course, you need to have clean water sitting around for this to work.
When you’re using a LifeStraw, you need yo keep in mind that the end that goes into the water can possibly be contaminated. Keep it out of any clean water you have unless you don’t care that you can contaminate it.
What kind of filter does LifeStraw use?
LifeStraws use a hollow filter microfiltration membrane style filter. Water is forced through the .2 micron pores of the filter as you suck up water.

The SteriPEN is a lightweight UV water purifier designed to quickly make water clean and safe for drinking. It eliminated bacteria and protozoa like most water filters, but it also kills viruses which filters can’t.

Does the SteriPEN really work?
The SteriPEN kills 99.9% of all bacteria, protozoa, and viruses. It either flat out kills or deactivates them which means that they can’t reproduce or make you sick.
This big difference is that the SteriPEN doesn’t filter anything out. You’ll deactivate or kill anything in the water, but you’re still going to drink it along with any sediment that’s in the water.
One of the best things that UV sterilization does (or doesn’t do) is not add any chemicals to your water. If this important to you, then you’re really going to like the SteriPEN!
How does UV light kill bacteria?
UV light is a type of radiation. UV-C, the type used to disinfect water, is normally blocked by the ozone but bulbs like the one in the SteriPEN produce UV-C because it’s so effective in stopping microorganisms.
When these microorganisms are hit by the UV light, it damages their DNA and stops them from being able to reproduce. This means that you can’t get sick from them because they need to be able to reproduce to make you sick. By reproducing in your body they eventually cause negative health effects.
Can you drink any water with the SteriPEN?
The SteriPEN kills or damages microorganisms. That’s it. This means that you can’t drink chemically contaminated water or water that has salt in it.
How long does a SteriPEN last?
The SteriPEN can disinfect 50 liters of water per charge and the bulb life lasts for about 8000 activations. 1 activation disinfects .5 liter and 2 activations disinfect 1 liter of water.
What kind of batteries does a SteriPEN use?
This SeriPEN has a rechargeable battery that comes as part of the unit. The USB charging port allows you to recharge the SteriPEN with any of your USB power packs or solar panels. This makes it great for me since I usually have a solar panel and battery pack with me.
You can find other versions of the SteriPEN that use AA batteries if you like batteries that can be replaced better. There are benefits of both so go with what works for you!
Can you use a SteriPEN in a metal bottle?
SteriPENs can be used in pretty much any container that you have. Glass, metal, plastic, etc. The container actually blocks any possible harmful UV light that gets produced.

Choosing between the LifeStraw and SteriPEN can be confusing but it’s actually pretty straight forward in my opinion.
Do you need to treat your water for viruses?
Most of the worst waterborne diseases that you’re going to find in North America are caused by either bacteria or protozoa. This means that you’re probably not going to need to kill off viruses in your drinking water. The one exception is the possibility of Hepatitis A.
Hepatitis A is spread when human feces from an infected person make it into a water supply. Hepatitis A is found all across the U.S. but it’s rare for it to be found in water supplies.
If you think you’re likely to contact water that’s contaminated by human waste, then the SteriPEN is what you want.
Most people will be better off with the LifeStraw
Anyone looking for an inexpensive filter that is easy to use and works well in cloudy water should go for the LifeStraw.
Conclusion
The LifeStraw and SteriPEN are two common pieces of equipment used to make potentially contaminated water sources clean and drinkable.
Both are small and pretty easy to use, but the similarities really end there. The LifeStraw is a personal water filter and the SteriPEN is a UV water sterilizer that can make drinking water for an individual or a group.
In the end, I’d suggest going for the cheaper LifeStraw over the SteriPEN unless you need to treat water for multiple people or you think you’re going to need to treat water for viruses.
