Ils se forment au fil des ans à partir de microcristaux de cholestérol qui apparaissent quand la bile est sursaturée en cholestérol et que la vésicule s'évacue mal. Guidage des vésicules

Le guidage des vésicules, c'est-à-dire le fait qu'elles arrivent à la bonne cible, se fait par la spécificité de récepteurs qu'elles ont à la membrane, les protéines RAB et SNARE notamment, qui interviennent à différents moments du guidage. Le schéma suivant reprend les étapes qui suivent la formation de la vésicule après son bourgeonnement.

Il y a d'abord une phase de migration le long du cytosquelette sur des rails de microtubules, à l'aide de protéines motrices comme la dynéine ou la kinésine selon le sens de déplacement (antérograde ou rétrograde). Ensuite la vésicule s'accroche à la membrane grâce à une interaction homophilique c'est-à-dire entre deux protéines de même type, RAB qui est sur la membrane de la vésicule et sur la membrane cible.

La phase d'accroche est à distinguer de la phase d'arrimage, qui fait intervenir les protéines SNARE. On en a de deux types : les t-SNARE (target) sont sur la membrane cible, et les v-SNARE (vesicle) sur la vésicule. Il y a deux t-SNARE pour une v-SNARE. Enfin, une fois arrimée, la membrane de la vésicule fusionne avec la membrane cible s'il y a un signal chimique, le signal calcique (Ca2+) qui permet donc la régulation de l'exocytose par exemple.

C'est un schéma simplifié et il existe en réalité bien plus d'acteurs au cours notamment de l'exocytose.

Ces schémas ont été faits pour mes ED du Tutorat à partir des cours que j'ai retranscrit quand j'étais en première année de médecine. Ma seule source est le professeur de l'époque, et je peux avoir mal compris certaines choses, faire des approximations fausses, etc même si je fais de mon mieux.
