I think they're "sisters" in the sense of brothers in arms, I believe. Carmilla does say that Vaggie was fighting like her "sisters", but I doubt it was a biological sense. It could also be like sisterhood/brotherhood, you know.

Considering Viv doesn't like incest, I doubt she would have Adam make a joke about how hot Charlie and Vaggie's relationship is, knowing that that was his "daughter".

Was Vaggie a human or did Adam created her?

Considering Adam is the one gave her that name, it's best to assume that she was never human. Not only that, but she also bleeds golden just like Lucifer. And, yes, Adam bled gold, but it's hard to say how much he is truly human, considering that he was made by angels.

Plus, I'm going off of Helluva Boss logic where it seems like hellborns bleed black, while Sinners bleed red.
