Enologist Emily Wiemer Is Honing in on Sonoma County’s ‘Yum Factor’

“Table fellowship” is what attracted Emily Wiemer, enologist at Three Sticks Winery to the wine industry. It’s sitting around with new people, friends, and colleagues learning about each with a wine bottle and cork as the only distraction is where she gains energy. The Minnesota native’s wine journey wasn’t too much of a stretch since she already had a passion for food and agriculture. She fully intended to pursue a job in the food and agricultural field after attaining an undergraduate degree in food science from Iowa State University. However, it was wine that “brought it all together” for her.
After completing an internship at Gloria Ferrer and her master’s degree in viticulture and enology at the University of California Davis, Wiemer joined Three Sticks in 2019. This year, the 2019 (Three Sticks current release) is her first “grape to bottle.” Wiemer sat down with VinePair to discuss and gain an understanding of the enologist role, her thoughts on women in the wine industry, and how she found the “yum factor” at Three Sticks.
1. Where did your passion for wine come from?
My passion for wine arose from the realization that it bridges many gaps among things I already love. Wine unites my love for agriculture that I cultivated while at Iowa State; it provides me with a medium through which to apply chemistry and microbiology; and it incorporates hospitality and sharing my passion with others. I also really appreciate that winemaking can be found in every corner of the globe. The pursuit of becoming a winemaker has allowed me to travel for work to South Africa and Italy. I love incorporating wine tastings into touring new places.
2. In your own words, what is an enologist; how is it different and/or related to winemaker?
The easiest way to put it is to say that I am a “wine scientist.” Much of my role involves sampling or testing wines to help monitor them. The data is then used in conjunction with tastings, but the response of our taste buds always wins out over a number on the page. Preparing and participating in tastings is another crucial part of my day-to-day role, especially during blending and bottling. I do lots of quality assurance and quality control, and through this I participate in every part of the winemaking process — from grape to bottle — which makes it a great preparatory role for moving into winemaking positions as I progress in my career.
3. There are several entry points into wine industry. What was yours and why did you choose it?
I entered the industry via education, knowing that it would be a powerful tool in helping me pivot and build upon my previous studies and experience in the food and agriculture industry. It was also an opportunity to begin building a community for myself in a new state and a new field. So, I enrolled at the University of California at Davis to learn more about the winemaking process and the science behind it. From there, gaining hands-on experience was crucial to cementing what I had learned in school and providing me a way to apply it.
4. How did UC Davis prepare you for your role and the wine world?
UC Davis helped me to further develop my critical thinking and data interpretation skills and helped me to build a network of wine industry professionals through the community of students, including my graduate program cohort. We were able to put these technical learnings into perspective through participation in extracurricular groups that also introduced us to industry veterans and a diverse array of successful approaches to winemaking. This helped us to realize how these lessons would best serve us, and to determine when we might want to take a different approach. Tasting groups, classes, and seminars helped each of us to discover our own personal style. In addition, these experiences helped us to acknowledge that the context of our schooling, particularly related to sensory perception, grape growing, and winemaking, can affect what we find in each bottle.
5. How did you end up at Three Sticks? In your opinion how does it stand out?
I knew as I entered the wine world that I wanted to focus on Sonoma County, particularly on Pinot Noir and Chardonnay, with quality at top of mind. Three Sticks is all of this and more. Carl Formaker, Three Sticks’ assistant winemaker (and a fellow UC Davis alum), helped to connect me with the rest of the winemaking team and it turned out to be a great fit. Three Sticks stands out because of our ability to get to know our estate vineyards across Sonoma County and what we on the winemaking team call the “yum factor.” We look for it as we’re blending, and we delight in it as we revisit wines we’ve made from previous vintages. It is this sort of X-factor that leaves you wanting another sip. I feel supported and encouraged, which has allowed me to thrive these last couple years. I’m so grateful for that.
6. What advice would you give to young women wanting to work in the industry as a winemaker, in the vineyard, or the cellar?
Just try it! And don’t overthink it! Be prepared for some hard work, but it comes with some great rewards. Find a team that supports you, and then “bloom and grow.” Try to learn why you’re doing a task — this helps both with motivation and fulfillment at work for me. Being humble, vulnerable, and ready to learn also fosters a longer-term commitment to crafting wines and remaining in this industry. Don’t let self-doubt or negative self-talk push you out of something that you really love.
7. How has the wine industry changed in the last year? How have things improved?
The past year was extremely challenging for the California wine world due to Covid and fires. These issues have forced us to adapt and improve. Having an online presence and the adoption of more technology were huge factors both for selling wine (online marketplaces, virtual tastings) as well as for increasing access for seminars and conferences while unable to gather or travel. Though these were imperfect replacements, I think that there were some highly positive takeaways that will remain in place. Fires forced everyone to confront their commitment to quality, though the response to this challenge varied across the industry. Some chose not to harvest, and others felt they could still make great wine since the effects were felt differently across each region. Regardless, we have all had to learn a lot more about this threat to our industry and how we can defend the quality of our wines against it.
8. What is the best piece of advice you received regarding your career?
Being reminded to balance patience with drive. While having goals is obviously very important to continue progressing, realizing the beauty of maximizing your current situation for the utmost growth has been a big lesson for me. Acknowledging that I am right where I need to be in the given moment has helped lend a lot more satisfaction and happiness to my journey in wine. Embracing this long road is crucial to promote sustainable learning for me.
9. How does the “affordability” of California affect interns, cellar masters, and other roles in Napa/Sonoma?
Because the area where we are employed is so highly coveted by others for its lifestyle, weather, and location, it can be tough to live where we work. I would say that it often ends up preventing some of us from participating in the “lifestyle” we had hoped for as we joined the industry, which is heartbreaking. But we enjoy other insider perks in wine country that help to offset this challenge, though they do not make up for the challenge completely.
Our hospitality colleagues warmly take care of their own, and that sort of acknowledgment is so appreciated. It can be tough for interns (particularly those that are traveling) to break even given the dual challenges of expensive housing and their temporary status. This then negatively affects their experience in exploring the region that they are working within.
10. Do you have advice for anyone interested in or new to working a harvest regarding expectations?
Be ready to get dirty and work long hours. It is extremely important to check one’s ego at the door, because saying “that’s not my job” is not OK. We are all in it together, and that camaraderie and teamwork is what buoys us through the vintage season. Seasonal work is a huge challenge as the hours and work are inconsistent as folks enter the industry, but we all patch it together until we find a sustainable, full-time job.
11. What’s next for you professionally?
I hope to keep growing in my role as enologist for Three Sticks and that I continue to learn from everyone on the rest of the team. Progressing into roles as assistant winemaker and eventually head winemaker will be stepping stones as I continue my career in wine. It is also important to me that I help as many people as possible as I grow and continue my learning. I aspire to do this by sharing my knowledge, offering encouragement, and by being inclusive in all that I do.
12. What’s next for Three Sticks?
We are continuing to strive to make the best wines possible. We will continue to make wines that represent their growing season and a sense of their place. This commitment means that we are never complacent or rocking back on our heels. It keeps us learning. It keeps us growing. It is also important to us that we keep sharing our wines, specifically in modes that convey table fellowship and that foster friendship and community. Gathering around our wines with friends and family is when they shine brightest. This organic growth is what will ensure and maintain our role in Sonoma County wine.

Terroir, Rootstock, Clones, and Cross-Breeding: Quality Wine Starts In the Vineyard

To the casual observer, wine might appear to be a relatively straight forward beverage to create. But now that I am wanting to plant a few vines the whole process is becoming mind-boggling; aside from the winery, just the simple task of planting a few vines is a series of complex considerations. Obviously, I am not purporting that my earlier wine making hobby efforts produced a quality wine; far from it. But I have concluded, just any grape juice, even in the hands of an accomplished winemaker, does not bring forth a quality wine. So, what makes a great juice?
I am reminded of an adage: Great wine starts in the vineyard. "The phrase is not meant literally but implies that you can't make great wine if the raw material isn't up to scratch (sic). This is certainly true and in the past two decades wineries have been putting a huge amount of effort into their vineyards in a bid to improve their fruit quality, trialing new methods from green harvesting to fancy irrigation systems," writes Rebecca Gibbs, Editor at Wine-Searcher.
There are indications that wines are being elevated starting in the vineyard. Just look at the amount of money being spent on new varietals, clones, new plantings, research into disease tolerant vines, and better soil chemistry/research. Even major universities are expending significant resources on research efforts that are improving the attributes/characteristics of wine grapes. For example, the University of Arkansas, under the direction of Dr. John Clark has a great wine grape program and has been awarded patents on brand new varietals. I am only pointing out that there is significant research, even in areas not known for being a bastion of wines. Of course, the recognized big players in wine research are: UC-Davis, Fresno State, Penn State, Cornell, Oregon, and Minnesota Universities.
There are many universities in the U.S. that have major programs underway to develop new clones, growing techniques and varietals that address specific needs of vineyard owners relative to improving plant and fruit quality. Such programs are bringing forward vastly improved fruit that make quality wines. The focus on grape/vine research is to improve the viticulture characteristics that address the ever-changing needs of the wine industry (and ultimately the consumer). With wineries in all 50 states, thus dictating differing needs of these geographically disparate wine centers, there is constant need for new ways to improve wine, starting in the vineyard. Consider this, a vineyard owner/winery owner may need new vines with distinct characteristics. For example: improved cluster size, higher yields, different fruit chemistry, color of the juice, phenolics, a specific aroma profile, drought and wet climate resistance, more disease tolerant, etc. Obviously, this is a challenge that only relates to vineyard operations. In an earlier article I mentioned that there is an ongoing research program with the Catena Winery in Mendoza, Argentina and UC-Davis to develop new clones (and maybe varietals) that grow quality grapes in high and arid soils and climatic conditions. Again, you need good fruit for good wine.
Probably the most recognized grape varietal for wine is Cabernet Sauvignon, a varietal from France that came about by an act of nature. There are references to this grape that goes back to the 17th century. It is a "cross" between Sauvignon Blanc and Cabernet Franc. This varietal is planted basically worldwide and has some very admirable character for winemakers; it is hearty, great color and nice aromas. (By-the-way, the ancestry of the Cabernet Sauvignon was validated by UC-Davis in 1997. "UC- Davis scientists John Bowers and Carole Meredith solved the mystery using DNA fingerprinting technology that proved that Cabernet Sauvignon was the progeny of a surprising spontaneous crossing of the Bordeaux cultivars, Cabernet franc and Sauvignon blanc," as reported in Foundation Plant Services 2008.)
With pollination happening in nature, even the Cabernet Sauvignon goes through mutational changes constantly. This would result in new clones coming about. Some have said there are 29 Cabernet Sauvignon clones.
Yes, we know vineyards select the vines they plant based upon a number of considerations, some were noted previously. In reality a vineyard manager, confronted with the need to plant new wines is confronted with a plethora of options for the vine specifications they will take to their vine nursery. The considerations run the gambit. For example, what are the soil conditions and what varietal will grow best? Climate trends can impact varietal selection and the clone type for that varietal. Then the vineyard manager must select a rootstock. And the list goes on. Sufficed to say: soil, climate, disease resistant, yields, colors and aromas of a grape can all be overruled by considerations dictated by the market place; what the consumer wants to buy. Sometimes the useful life of a vine (vines can produce for approximately 75 years) is never achieved because of considerations outside of human control.
So, when you are driving through vineyards in Sonoma or Napa on the way to a wine tasting, let's explore the options that go into producing a high-quality fruit that will make high quality wines. All vineyards are not created equal. Today we know that the wine consumer is paying considerably more for quality wines. Ultimately, the vineyard owner will select a vine that will produce fruit that a winery will buy, that will also meet their specifications. After all, there are more than 10,000 varieties in the world. Of course, there are probably only a thousand or so used for wine and there are thousands of clones within varietals.

Disease, injury, and other stress factors harm cells throughout our bodies. Ideally, the damaged cells are cleared by our immune systems through a process called apoptosis. But as we age, our bodies are no longer as effective at removing dysfunctional cells, and this can contribute to a weakened immune system and other less efficient biological processes.
An increasing number of researchers are exploring whether learning to harness a cellular state known as senescence — during which damaged cells resist removal by apoptosis, linger, and harm neighboring normal cells — might hold the key to revitalizing aging tissues and increasing healthy, active years of life.
What is cellular senescence?
Senescent cells are unique in that they eventually stop multiplying but don’t die off when they should. They instead remain and continue to release chemicals that can trigger inflammation. Like the one moldy piece of fruit that corrupts the entire bowl, a relatively small number of senescent cells can persist and spread inflammation that can damage neighboring cells.
However, not all senescent cells are bad. The molecules and compounds expressed by senescent cells (known as the senescent secretome) play important roles across the lifespan, including in embryonic development, childbirth, and wound healing.
How cellular senescence affects the body
The number of senescent cells in a person’s body increases with age. As the aging immune system becomes less efficient, senescent cells accumulate and taint healthy cells. This can affect a person’s ability to withstand stress or illness; recuperate from injuries; and learn new things, since senescent cells in the brain can degrade cognitive functions.
As a result, cellular senescence has been connected to a multitude of age-related conditions, including cancer, diabetes, osteoporosis, cardiovascular disease, stroke, Alzheimer’s disease and related dementias, and osteoarthritis. It has also been linked to declines in eyesight, mobility, and thinking ability. Investigations are underway to see if senescent skin cells may contribute to sagging and wrinkling, and if senescent cells might also be connected to the cytokine storm of inflammation that makes COVID-19 so deadly for older adults.
A scientific curiosity
Cellular senescence has been on scientists’ radar since the early 1960s when Leonard Hayflick, Ph.D., and his colleague Paul Moorhead, Ph.D., overturned the long-held scientific consensus that human cell samples could replicate endlessly in lab cultures. Hayflick and Moorhead showed that there was a limit on the number of division cycles, after which cells entered senescence.
For long after that finding, senescence was believed to be just an odd side effect of laboratory cell culture environments. It was poorly understood and studied by only a few research teams, but in the past 20 years, there has been a spike in interest. Today, it is a still young but promising scientific discipline that has sparked more NIH research as well as private industry support for studies to discover and develop drugs that might give Mother Nature a boost in clearing out senescent cells.
A pioneering pursuit to extend health span
Jim Kirkland, M.D., Ph.D., of the Mayo Clinic, and his former colleague Jan van Deursen, Ph.D., were pioneers of the senescence renaissance. For nearly two decades, Kirkland has studied ways to remove senescent cells. A clinical geriatrician, Kirkland often says he grew tired of prescribing the latest innovations in wheelchairs, walkers, or incontinence control. Instead, he wanted to learn if it was possible to slow down or partially reverse the fundamental aging processes in humans that lead to common health issues as we age.
Kirkland and his team currently focus primarily on a cocktail of two drugs: dasatinib (D), a drug commonly used in leukemia chemotherapy; and quercetin (Q), a pigment found in strawberries, grapes, tomatoes, red wine, onions, and other fruits and vegetables that has natural anti-inflammatory properties. When administered together, D&Q function as senolytics, i.e., drugs that clear out senescent cells.
In 2019 Kirkland and colleagues conducted a very small pilot study of D&Q in 14 volunteers who had idiopathic pulmonary fibrosis (IPF), a fatal, tough-to-treat, and debilitating lung disease. The results showed that the senolytic combination improved physical function in participants, demonstrating the feasibility of testing D&Q for this condition in larger controlled clinical trials. In subsequent small clinical trials, the Mayo Clinic team found that D&Q also cleared senescent cells in research participants with diabetic kidney disease.
In mouse models of IPF, D&Q was shown to clear senescent lung cells, reduce inflammation, and extend health span (the years of life free of major disease and disability), but not longevity. Researchers are currently exploring the potential of senolytics for osteoporosis, glaucoma, macular degeneration, diabetic neuropathy, and other age-related conditions.
Kirkland’s lab has shown that middle-aged mice given a variety of senolytics had delayed onset of several age-related ailments compared to untreated peers. Older mice given D&Q were faster, stronger, and spryer than control groups, and the positive effects lasted until the final months of their natural lifespans. Other studies showed that older mice given D&Q had a 36% longer average lifespan than untreated peers.
Kirkland’s team has also tested the opposite approach, by injecting healthy young or middle-aged mice with a customized senescent cell type. This intervention caused rapid deterioration in their mobility, speed, and strength, and big jumps in frailty rates. What’s more, these negative effects lingered long after the transplanted senescent cells had died off.
A long way to go before safe human use
While these studies pose exciting scientific and medical questions, Kirkland is adamant in underscoring the huge gap between mice and humans. He regularly urges people not to take senolytics or similar supplements or drugs outside of a clinical trial because they have not been confirmed as safe.
“We haven’t so far seen serious or severe adverse events in clinical trials with some of these agents, but it doesn’t mean they won’t happen,” said Kirkland. “They could take a long time to develop, and anything that sounds too good to be true in mouse or cell culture models usually is. We need many more long-term studies and human trials.”
Kirkland, his Mayo colleague Tamar Tchkonia, Ph.D.; and Stefan Tullius, M.D., Ph.D., of Harvard University Medical School, are also looking at other potential benefits of taming senescence to rejuvenate older tissues. One such project is exploring whether treating kidneys or livers from older organ donors with senolytics prior to transplant could help repair the damage accumulated over time and with age. If true, this could make older organs more viable and safer for transplantation, and thus reduce waitlists.
Kirkland emphasized that there are many connections between senescence and age-related conditions, making the field ripe for future discoveries.
Growing interest nationwide
NIA-supported scientists working at NIH and throughout our nation continue to explore the mechanisms that regulate the intersection of senescence, inflammation, aging, and disease. Chronic inflammation seems to be a key to how senescence can turn from helpful to harmful. Healthy tissues normally have a limited type of senescence that suppresses abnormal cell growth that could lead to cancer. But when the healthy cell environment is disrupted by injury or age-related inflammation, this can trigger a loss of control of the body’s natural regulation of senescent cells, which may be linked to the development of cancer and other diseases.
Two top experts in the burgeoning field and frequent collaborators with Kirkland are University of Minnesota researchers Laura Niedernhofer, M.D., Ph.D., and Paul Robbins, Ph.D. Niedernhofer came to senescence from a background in DNA damage and repair.
“Senescence is fascinating to me because a damaged senescent cell can in turn cause damage in healthy cells,” she said, stating further that normal senescence can be helpful, as in wound healing, and that senescence itself is a potent tumor suppressor mechanism.
“Picture a wound on your arm. There is growing evidence that senescent cells may be critical for calling in immune cells to help heal that. But it’s just a temporary thing to close that wound and then the signal should go away. The problem comes when you’re not able to clear those senescent cells. They are great at spurring the immune system to action, but when your immune system falters as you get older, you get these chronic senescent cells.”
Robbins quipped about the inevitable rise in senescence as we age.
I think I’m chock full of bad senescent cells. They start accumulating when you hit age 60, and from there it’s kind of an exponential increase. Toward the end of life, depending how you’re aging, up to 10% of certain tissues can have at least some markers of senescence.

Senescence science collaboration across NIH
NIH-wide interest in how cellular senescence factors into multiple diseases and conditions is growing as well. The NIH Cellular Senescence Network (SenNet), organized by NIA and the National Cancer Institute, is an NIH Common Fund initiative to coordinate the exploration, identification, and cataloging of differences in senescent cells throughout the body. The network also supports research collaboration and data sharing that could lead to future therapeutics. Through SenNet, researchers aim to create a four-dimensional atlas of senescent cells involved in healthy human aging to find and characterize healthy and unhealthy senescent cells. This could inform investigations into how future senolytic therapies can be customized to only target harmful senescence.
Robbins, Niedernhofer, and their colleagues are energized by the future possibilities.
“Before you go in for surgery, you might want to clear your bad senescent cells to help you recover faster,” Robbins said. “Cancer radiation or chemotherapy treatment at a young age has been shown to drive senescence and aging, so there’s a whole group of cancer survivors that may be very appropriate for this sort of treatment.”
Niedernhofer added that the senescence field has exploded since she and others described the first senolytic compounds in 2015.
Now there are more than 20 clinical trials registered for anything from chronic kidney disease or pulmonary fibrosis to frailty in nursing home patients. It’s moved very quickly and the thing to be proud of is, it’s really been driven by NIH-funded investigators.

Sorting out good senescence from bad
Other NIA-funded researchers, like Judy Campisi, Ph.D., of the Buck Institute, see senolytics as a potential routine treatment of the future that could help older adults stave off age-related illness and frailty longer, similar to a dental check-up. In the San Francisco Bay area where she lives, multiple public and private research institutions are looking into future medical applications for senolytics.
Campisi began her career focused on how senescence affected cancer. Eventually, she expanded her research into how it contributes to aging and age-related disease.
“We made a mouse model which allowed us to eliminate senescent cells throughout the lifespan, and it improved a lot of age-related diseases in mice,” she said. “Now the question is, could this happen in humans using senolytics? Well, it does in human cells and in certain human tissues.”
Campisi and her collaborators continue to investigate various aspects and mechanisms of senescence, including how it impacts osteoarthritis and sarcopenia (loss of muscle mass later in life that can lead to disability, frailty, and fall risk) and if senolytic treatments can help reduce the harmful side effects of chemotherapy and HIV/AIDS drugs.
Campisi concedes there are still many unanswered questions.
“We know there are good things senescent cells do in healing, tissue repair, embryonic development, and childbirth. But how are the good guys different from the bad guys? And is there a way we kill the bad guys and not the good guys? We honestly don’t know that yet.”
Mapping senescence targets across the body
Myriam Gorospe, Ph.D., and her colleagues in NIA’s in-house labs are examining whether senolytics could tamp down harmful senescence in lab and mice models of Alzheimer’s disease linked to problems with thinking and cognition. They are hoping to map and manipulate senescence-related proteins that are involved with the buildup of beta-amyloid plaques and neuroinflammation, hallmarks of Alzheimer’s disease.

Gorospe’s lab is pursuing the quest for biomarkers — measurable substances in the body that indicate the presence of disease — to better pinpoint and target senescent cells in different types of tissue. Her team also works with senomorphics, a different category of drugs that don’t eliminate senescent cells, but instead repress their harmful secretions. They study senescence’s impact on aging body tissues like the lungs and muscles, and its role in conditions like arteriosclerosis and Alzheimer’s-related neurological decline.
Scientists are just beginning to take a closer look at the role senescence plays in one of the most serious public health crises in recent history, the COVID-19 global pandemic.
“Some of the biomarkers on the membrane of senescent cells function as receptors for the SARS-CoV viruses, so they’re critical in understanding the acute pathology of COVID-19 infection,” said Gorospe.
Lessons learned from the pandemic could yield useful knowledge as the scientific community moves forward to someday testing if human use of senomorphics or senolytic treatments could help extend healthy lifespan and ward off disease.
Cautious optimism, support for a young science
In August 2019, NIA convened a workshop attended by academic, industry, and public health scientists to study if it was possible to safely repurpose some drugs or dietary supplements for their senolytic or senomorphic effects to treat or prevent age-related conditions. NIA and the broader NIH offer growing funding opportunities in the field through SenNet and other efforts. Currently, NIA supports a growing portfolio of extramural senescence research, coordinated by program officers including Viviana Perez Montes, Ph.D.
Perez Montes and her NIA colleagues agree the potential for harnessing senescence is vast but echo the cautions of Kirkland that this field is still very young, with a lot of work yet to be done, including building a common research infrastructure and safety standards for human studies.
Above all, it’s important to manage expectations for the public and the medical community that any safe potential health benefits from manipulating senescence will be years if not decades in the future.
“We have to remember that senolytics are going to kill senescent cells and we believe that some of those senescence functions could be beneficial,” said Perez Montes. “If you kill and eliminate cells that are not going to be replaced, we don’t know yet the biological and physiological consequences of this elimination, so we need to be extremely careful in future translational and clinical studies because we’re still in the very early stages.”
