Ughhhhh...
I do like to think that after the events of Desperados 3 and its DLC, Cooper's gang split — mostly to avoid being such a prominent target for the law enforcement. John and Kate then went to meet Sam on their Santa Fe job, and Doc and Isabelle went on their own jobs.
Won't lie, I'm tempted to write a drabble where the latter two successfully pull a number of jobs... before biting more than they can chew and an informant betrays them. McCoy gets captured and gets his eye shot out point-blank by his own Buntline, and Isabelle has to give up the location of their stash before Doc's kidnappers shoot out his other eye. Cue a "let's see you snipe with your eyes gone, bastard" snide comment from the kidnappers. Isabelle outsmarts and kills them all. She gets Doc out and, with the money, both flee into the wilderness.
The rest of the story would be Isabelle nursing Doc back to health (she is a voodoo witch after all) and McCoy having a bit of existential crisis as he thinks he won't be able to do his job anymore i.e. be a sniper. Plus, losing an eye really, really tests his "nothing personal" attitude — he's, understandably, overcome with Emotions (TM).
Doc decides to take it slow (call it his convalescence) and Isabelle lets him use her cabin deep in the bayou. With time and practice he regains his sniping skills, but Isabelle, unfortunately, is found and lynched by the company guys from before.

Hello, I've heard that you're an expert on LOS lore. Some people claim that in one trailer for LOS2 you can see Dracul visiting the tombs of Marie and Laura, do you know anything about that?
Oh, with the advent of Netflix Castlevania, I've more or less left the LoS fandom... But I can tell you that those tombs were just a set piece; Gabriel never mentions them anywhere in the script.
