 Usage Method of Liquid Chromogenic Culture Media
1. Container surface sterilization: The surface of the aluminum foil should be disinfected with an alcohol lamp before use.
2. Opening: Open the aluminum foil with an MT opener (OS-01) or open directly with a sterile pipette.
3. Inject the sample diluent: use a pipette to inject the sample dilution into the cup
XM-30: 1ml, XM-31: 10ml, XM-32: 5ml
4. Cultivation: Culture time: 24 hours; Culture temperature: 36 ± 1 °C
5. Disposal: Dispose of them after sterilization
※XM powder medium usage: Dissolve 500ml of pure water per bag, inject 10ml into the test tube, autoclave, then inject 1ml sample, and incubate at 36±1°C for 24 hours. The identification method is the same.
 Coloring Principle
Coliforms positive
There is a special β-galactosidase only exit in the coliforms. It can be used as an indicator enzyme for coliform bacteria, which catalyzes the hydrolysis of the substrate X-GAL to form 5,5-dibromo -4,4-dichloro indigo, appearing light blue ~ purple-blue.
E. coli positive
β--glucuronidase can be used as an indicator enzyme for Escherichia coli; because β-glucuronidase can be observed in Escherichia coli, Salmonella and Shigella in intestinal bacteria, and 95% of E. coli contains β-glucuronidase. It catalyzes the hydrolysis of MUG to form 4-methylumbelliferon, which exhibits fluorescence at 366 nm ultraviolet light and is visually discernible under indoor light intensity.

Diluents Series
In daily practical experiments, when the liquid samples are collected correctly, we often do not need a pure solution to carry out experiments. In order to get better and more accurate detection results, diluent water or other solutions will be added to obtain the solution with lower concentration. In this process, the solvent diluent is needed. In many laboratories, it is generally believed that direct preparation of buffer by diluent reagent will be more accurate and cheaper, which is not in line with the fact, because the quality of buffer is related to the purity of reagent, the accuracy of reagent weighing, the quality of purified water and other factors such as temperature, preparation container and so on.
   ELEX diluents series has greatly improved the trouble and inaccuracy caused by the preparation of diluents on site. The product mainly contains two types of diluents: gradient dilution and sterile diluents.
 Gradient dilution is the most widely used in the laboratory. It has phosphate-buffered saline, 1% buffered peptone water and 0.1% phosphate-buffered peptone saline. Other solutions can be customized according to customer needs. The wide push plate design of the front end of the gradient dilution liquid bottle cap minimizes the possibility of contact pollution when opening the cap; the tongue pressing design of the back end of the bottle cap facilitates the easy control of the opening angle of the cap by one hand, which make the operation more convenient, reasonable and convenient.
 Sterile diluents mainly have MT-11 series, MV2-90 series, MV-225 series (wide mouth), MV2-1000 series and MV-1000 series, equipped with PBS, BPW, MRD and AP solutions. MT-11 series contains 9ml diluents, which is similar to gradient diluents GD-9 for gradient dilution. Customers can customize different types of diluents according to their own needs. Make food microbiological testing much easier and more effective
Compared with the agar medium, our bacterial culture media plate makes the detection of food microorganisms easier, more standardized, and more efficient, but without pollution.
1. Culture media ready to be used after opening;

Notes
1) The product is used for the microbiological examination of foods and beverages, and cannot be used clinically.
2) Do not open the tectorial membrane before transplanting the sample.
3) Do not use expired products.
4) Do not use products that are damaged, deformed, discolored, dirty, or foreign matters.
5) Do not expose the product to ultraviolet light or sunlight.
6) Do not press the film after the sample liquid is dropped. This will cause the sample solution to overflow to the outside of the culture area.
7) When the sample solution overflows from the culture area, replace it with a new one.
8) If the product enters the eye or mouth, immediately wash it with water and go to the hospital.
9) When using the product, it is susceptible to microbial contamination. Perform specific operations under the guidance of relevant qualified personnel.
10) Please note that the sample or the product that has been in contact with the sample liquid is a contaminated item.
Preservation method
Keep in cold storage (2~8°C), fold it twice at the opening of the bag after breaking, and fix it with tape.
Validity period
The validity period of the product is written on the aluminum bag and the upper part of the product (the date described after "EXP" is the validity period). Please use the aluminum bag within 3 months after opening.
 Abandonment method
The product has a risk of secondary pollution after use. Therefore, after proper sterilization, it should be disposed of according to the waste standards of the respective samples and related facilities.
Guarantee responsibility
When the product defect is obvious, replace the corresponding quantity with the new product. The judgment and application of the inspection results are the user’s responsibility, and the manufacturing company and the agent of the product are exempt from liability.

In order to get better and more accurate detection results, diluent water or other solutions will be added to obtain the solution with lower concentration. In this process, diluents are needed. In many laboratories, it is generally believed that direct preparation of buffer by reagent will be more accurate and cheaper, which is not in line with the fact, because the quality of buffer is related to the purity of reagent, the accuracy of reagent weighing, the quality of purified water and other factors such as temperature, preparation container and so on.

Culture Media Series
In microbial detection, when the sample is collected and diluted correctly, the prepared sample solution needs to select the appropriate culture medium for cultivation, so as to judge whether the sample meets the standard. The choice of culture medium means whether we can get the desired experimental results quickly. ELEX is a culture media supplier providing culture medium that is mainly used for rapid detection, to make the microbial detection more simple and effective. The products mainly include DNP Medi·Ca culture medium plate, concentrated liquid chromogenic medium, tri-color agar medium, and liquid chromogenic medium.
Different types of culture media for bacterial growth have their own characteristics. Compared with other agar media, DNP media can make the food microbial detection much easier, more standardized, more efficient and pollution-free. Compared with the traditional medium, Medi·Ca culture medium plate has the following advantages. Firstly, the preparation process of the medium is omitted which saves time; the steps are reduced which lower the possibility of contamination in the process. Secondly, the medium plate can be placed overlapping, saving about 95% of the cultivation space, in addition to reducing emissions of exhaust gas and greenhouse gas (GHG), which save s environmental protection costs. Thirdly, it is easy to operate and does not depend on the operator's level. The bacterial colony is easy to see with convenient judgment and accurate result.
Compared with other similar products, it also has the following advantages: firstly, sample dropping can be carried out uniformly without pressure plate with fewer steps which is fast and convenient; secondly, there is a special anti-spillover ring, sample liquid will not spill and lead to cross-contamination.
Concentrated liquid chromogenic medium ST-SSP can be used in combination with our swab simpler, and can be used for the qualitative detection of E. coli and coliforms at the same time.
Tri-color agar medium is widely used for simultaneous qualitative and quantitative detection of coliform bacteria, Escherichia coli and other intestinal bacteria in food, water, milk, ice cream and meat products. Colonies show three colors, which can be easily judged and counted.
There are two kinds of chromogenic substances in liquid chromogenic medium, which can directly identify coliforms and Escherichia coli, but only for qualitative detection.
