I think the relationship between the queen and the Scarecrow is a kind of Queerplatonic, while the queen and the king are family members who have feelings after a political marriage

In fact I had always thought the King was gay .Because he clearly preferred men, the only person he trusted is the king's hand and he built a statue of himself with him, calling the alchemist and the royal gardener “best friends”. But he never mentions the Queen, doesn't have much contact with the TimeKeeper, and even treats the corpse of the Tutorial Knight very roughly, though he treats the corpse of the royal gardener much more gently

The Scarecrow (aka the Royal Gardener) is asexual, as he doesn't seem to be romantically attracted to the other characters. But his relationship with the Queen and the king was so close that he even wrote for the king (red roses and blue violets), and was angry that the alchemist was called “best friend” by the king, or that he missed the Queen at the end of his life and wished to see her, and his words also show that he and the Queen were close to each other beyond ordinary friends. It is therefore highly likely that he is asexual, plus he was once a human (as different to non-human giant, alchemist, and timekeeper) and he is not married, although it appears that he is about the same age as the King, but the king already in fear of dying of old age

I can't say what's the Sexual orientation with the Queen, because she's not even human. Given that her character design is related to Athena (who is represented by the Queen of Spades in the game's achievements, who also owns a temple, and she as a serpent, and has owls that guards the castle), she may simply not be interested in humans, and not feel any sexual or romantic attraction to those currently appearing. But I think she developed a Queerplatonic relationship with the Scarecrow because she loved the scarecrow, she put mushroom boi plush that he sewed by hand in her bed to sleep with it, and she loved roses (and it turns out in boss rush that roses are the symbol of the scarecrow, and the symbol of the queen is the jewel on her chest).
